Mapquest has a spot where you can get the to and from in miles and put in your mileage and it will tell you how much fuel will cost depending on that.

from san diego its like 4.5 to 5.5 hours so in a v8 that would relate to 3/4 of a tank or so. from everything I have heard or seen that place is pretty awesome, one of those do it once type of things.

When i take my truck to pismo (01 Chevy 1500) it takes a full take to get there and back, useually about $60-90. Just remember a 4x4 is highly recommened at pismo and cars really arnt a good idea. I go there several times a year, well worth the drive.

I lived next to pismo from '95-'03 and went out to the dunes to either ride rented quads or just camp at the beach in front of the dunes staging area. The place is a blast and is well worth more then just one trip.
As far as a type of vehicle, a 4x4 really is best, but not always needed. Basically coupled with the fact that you are driving down the beach for several miles (stay in the tracked areas or down by the damp sand) there is a small river that has to be crossed to get to the camping/dune staging area.
Depending on the amount of rain the area has had recently. That river can be a trickle to a several foot deep torrent. I've seen lifted 4x4 trucks pushed out into the tides by it, i actually have a friend that lost her car to it (doesnt happen often, but it does).
There are usually several rigs waiting for people to get stuck in the water or sand, then they charge to pull you out.
All that being said, ive crossed that river in a '86 taurus station wagon, ford EXp, celica's etc. Just be mindful of the weather for your trip.
A link to some forums showing pics of the river-
