Carb Problem

I have a 1995 timberwolf yfb 250 of witch i paid $300 for and since then i have rebuilt the engine compleatly . I cleaned the carb and it will only run on full chock and when you try to run it without the choke it misses and backfires. Could some one out there give me some advice on this
