My 400 will take one, but it's not stock,,,to me (excluding the 05 Z400's, cause i dunno) they are not as comfortable and predictable as my EX,,,i like the fact that they are liquid cooled and reverse (now the 05 EX have rev) and parts are plentiful,,,they are very close stock for stock, but now the Z/KFX/DVX have more potential than the EX's, but i wouldn't get rid on mine for one.
O, to answer your Q's, they make good power, respond well to mods, operate easily. I would expect that they would not fair well against an R of any kind, My X would be a much fairer drag race.

Everyone who has ridden my 05 KFX400 has been extremely impressed, even 660R and YFZ riders. I've read somewhere that the 05 is quicker than the 03-04 models.

Goto 350XRiders.com and click on the 350X VS Z400 link to get an idea of how fast they are. (Same machine, but not green) Now, keep in mind the trike rider got about as much outta that old dog as possible. The X was and still is 100% stock in the performance dept, and the Z had some minor mods...exhaust, airbox and jetting. Look at some of the other clips too. No more of the Z, but you can see how the X compares to a new 450 Yammy.
I think the Z/Kawi are great all around machines, and with some $$$$ spent on the RIGHT things, it can be a VERY quick ride. There is one Kawasaki (490cc
SERIOUSLY MODIFIED) that drags around here and it basically stomps the SHITE outta most everything...including moderately modded Shees.
